文拉法辛可以治疗偏头痛。根据2019年《美国头痛协会关于将新的偏头痛治疗纳入临床实践的立场声明》，文拉法辛属于偏头痛预防性药物治疗中有效的一类药物[1]。

文拉法辛是5-羟色胺和去甲肾上腺素再摄取的双重抑制剂，其主要的不良反应包括恶心、便秘、厌食、头晕、口干、热不耐受和出汗过多等[2]。在一项随机安慰剂对照试验中，每日使用150mg文拉法辛的试验组每月偏头痛发作的天数要显著少于安慰剂组[3]。另一项比较文拉法辛与阿米替林疗效的试验显示，两种药物疗效无显著差异，但文拉法辛不良反应的发生率要明显低于阿米替林[4]。在2009年的《欧洲神经病学学会偏头痛药物治疗指南》中，文拉法辛被推荐作为偏头痛预防性治疗中第二选择的抗抑郁药物[5]。

目前，多国用药指南在预防性偏头痛治疗中虽有提及文拉法辛，但仍需要更多高质量的临床随机安慰剂对照双盲试验来验证其疗效[1, 5]。
